The Unique Tutoplast Process

The Puros Allograft family of products is refined using the proprietary, 5-stage Tutoplast sterilization process. While preserving the valuable collagen matrix and tissue integrity, this process gently removes unwanted materials such as cells, antigens and viruses, as well as inactivating pathogens. Tutoplast-Processed tissues have been used for over 30 years in more than one million procedures without a single reported case of disease transmission.

  1. Osmotic treatment – Kills bacteria, destroys and removes unwanted cells, further reduces viral load
  2. Oxidative treatment – Destroys remaining proteins and minimized potential graft rejection
  3. Alkaline treatment – Further reduces prion infectivity
  4. Solvent dehydration – Removes water from tissue and acts as a disinfectant, further reducing prions
  5. Limited-dose gamma irradiation – Provides a sterility level of 10-6 and preserves graft integrity

Is the Tutoplast process different for soft tissue and hard tissue?

There is one slight difference between the hard and soft tissue processing. Hard tissue processing utilizes a delipidization step to inactivate lipid-enveloped viruses (such as HIV and HCV) and to remove fat, enhancing the healing characteristics. Soft tissue (pericardium) processing mechanically removes fat and includes an alkaline treatment of 1N sodium hydroxide (NaOH), at room temperature, for one hour. This processing step is scientifically recognized as an acceptable and effective methodology for reducing prion infectivity. Tutoplast processing has been shown to reduce the viral load of tissues by 12 logs, which is 10,000 times higher than any viral load ever recorded in a human patient.

The Tutoplast® Process has been fully validated for preservation of tissue biomechanical properties, viral inactivation, prion inactivation, healing and elimination of antigenicity.
